This is about Sonic Hedgehog Protein - it what was in one of the issues

in 2003 of CMT...Today magazine. Just a bit of a change since - Curis

has a new partnership with Wyeth. Elan is out of the picture.

Within each nerve, there is a signaling pathway called hedgehog. (this

has nothing to do with the hedgehog animal!) When the hedgehog pathway

is insufficient, nerve regeneration is slowed or absent. The hedgehog

pathway also regulates tissue and organ formation throughout human

development and adulthood. The signaling pathway of Hedgehog (Hh) that

occurs during development is often the same pathway the body calls on in

adulthood to maintain physiological balance and to repair and regenerate

nerves.

Curis is a therapeutic drug development company with a technological

discovery on regulating the hedgehog pathway that controls and repairs

nerve regeneration. One Curis product involves using proteins or small

molecules to modulate this pathway. This is sonic hedgehog protein.

The Hedgehog (Hh) pathway is essential for the formation of normal

nerves in the peripheral nervous system, as well as in the central

nervous system. Curis scientists have shown that treatment with sonic

hedgehog protein accelerates nerve recovery in animal models of nerve

trauma and disease. This protein has also proved successful in human

trials of diabetic neuropathy.

Curis scientists have developed a series of small molecule Hh compounds

that mimic the activity of the protein. These compounds could be taken

in pill form. Some of these small molecule Hh compounds are more potent

than the original Hh protein. These compounds are being evaluated

further before human trials begin for peripheral neuropathies, including

HMSN/CMT.

Hedgehog protein research is now being conducted under a joint venture
