Hi Mandy--

My husband has been on Sprycel for about 6 months. First couple weeks were a

little odd with strange rashes, some swelling around the eyes, fatigue and some

headaches. I say " odd " only because sometimes we would be talking, I'd go to the

bathroom really quick and come back and all of the sudden his entire neck would

be rashed! So the side affects literally could come out of the blue and last for

an hour or longer and then go away as fast as they came on. But all in all his

experience on Sprycel has been way better than on Gleevec. In fact, I think his

favorite thing is the fact that on Sprycel he can take it with or without food

and it is fine either way (I think they recommend taking it with food, I am just

stating what 's experience has been!. On Gleevec, he had to gourge himself

before taking it and most days he would still get extreme fatigue for about an

hour or two after taking it, not to mention a very upset stomach.

All in all other than what I have mentioned above, he has been pretty " normal "

on Sprycel. He is at the higher 140mg doseage (taken twice a day), and has

pretty much no side affects. Well, he does require more sleep than he used

to--at least 8-9 hours per night, but other than that is doing very well. Our

" new life " involves earlier bed times, less drinking, less physical activity

like snowboarding or working all day in our yard, but we always wonder if that

is due to our getting older and the fact that we now have a 1 1/2 year old!

Those little guys should come with some warning labels: toddlers may cause:

severe fatigue, headaches, muscle pain (from picking them up all the time!),

upset stomach (from the nasty diapers they have!), and the list goes on!! :)

My husband went off Gleevec due to liver toxicity...may I ask why your husband

is switching?

Hi,

First of all I wish you well, My experience is the that I had severe migraines

for about 3 weeks. Some soft stool, but that was it. I took tramadol 100mg for

the migraines, since tylenol and advil, aleve were all contraindicated by the

doc. Well good luck on your journey.

Hi Mandy,or the first 1 1/2 years I didn't have any side effects at all. Now,

for the last 3 weeks, a.\ll 10 finger tips are numb. It's getting better but

still there. Minor compared to what it could be, don't you think?

I've mentioned this before---I had a serious reaction to Sprycel which affects a

very small percentage. It was shortness of breath followed by pulmonary edema

that sent me to the ER and ICU. It came on quite suddenly and was so severe that

I had no doubt that I needed to get to the hospital. I'm back on Gleevec (400mg)

and am doing better on a reduced maintance dose and have milder side effects.

Good luck. I hope Sprycel works better for others. It sure would be nice not

to have a puffy face and weight gain.

Thanks to all who have already responded. DH was on 600mg Gleevec

since diagnosis in 2/07, but stopped responding to Gleevec in the

last month and moved into a blast crisis. The doctor tested his

Gleevec level (thanks to a free test from Novartis) and said there

was plenty of medicine floating around in there, but his CML was

resisting it. Which is really too bad because he had very minimal

side effects on the drug and was able to work and exercise.

We are adjusting to the Sprycel and likelihood of a transplant. It's

a lot to wrap our heads around, but I'm sure we'll get there. DH has

mentioned a very itchy head several times and worries about hair loss

with the new meds. He's also gotten rash like symptoms -- bumpy

skin, although not really itchy.

He's only been on Sprycel about a week, but it's so much nicer to be

educated about potential symptoms so that we are reassured if we see

them.

Yes--scratchy head..forgot to mention that one. Yep-- had that, too...lasted

about 2-3 weeks then went away.

Good luck on your CML journey, hope Sprycel gives him more success than Gleevec.

my daughter is on sprycel and the doctor told her she can take tylenol extra

stringth, I would like to know who told you MEl that tylenol were not good with

this pills, she have been taking tylenol like crazy , because of headach,

please let me know more info, im very nervous about this, i woudent want some

more bad reaction to her meds , she had a ruff time with gleevec, and she just

started on sprycel, and i woudent want some thing els to go wrong, please let me

know , so i can educate miself and her , thanks

I had severe headaches (burning worst than a migraine if possible) almost from

beginning. Lasting anywhere from 3 seconds to 5 minutes and somtimes continue

for a couple of hours and then went away eventually, havent had one months.

Joint pain fatigue. If you really excert yourself the muscles burn almost scream

at you. But all is something you can live with. I have been on Sprycel since

April to now.

I had some hair loss but not real bad, but has stopped and is coming back, yes I

too have the numbness in my fingers and toes but not as often as it was. Dry

skin has become the major problem now. And red, itchy splotches on my face, but

am waiting for it to go away as the other things have with time. I drink almost

a gallon of water a day that keeps the stomach cramps to a minimum. But not

every one has the same side effects. Still much better than the experiences of

Gleevic.

I thought that it was a no-no to take tyleonol while on Gleevec because of the

fact that Gleevec is processed by the liver, and so is Tyleonol and the thinking

was that one should not overload the liver. My husband had to go off Gleevec due

to liver toxicity, and so when he switched to Sprycel there was alot of talk

(with his oncologist and Dr. Druker), about how most likely he would NOT have

the same liver issues on Sprycel because it is not processed by the liver. So,

because Sprycel is not hard on the liver, it makes sense that one could take

Tylenol. BUT--yes--conflicting opinions...and this is just MY thinking on the

issue--so take it with a grain of salt I guess. :)

I was told by my Onc and also by the lead trail specialist at MDACC orlando. It

has a liver effect, and so does any tyrosine kinase inhibitor, so they said no,

no.

That is my experience.

It may depend on each case. I am on gleevec (400mg, since 2005), my

liver numbers were never abnormal and my hemato let me take tylenol,

in moderation. I don't need it often. When I have a cold I take 1 or 2

daily pills of sudafed. Some tylenol also when I get some joint pain

after climbing (maybe some damage from when my wbc was sky high, or

just getting older). I alternate with aspirin or ibuprophen when

necessary (what happened only once and for just 2/3 h since 2005).

if she cant take tylenol what should she take with sprycel, help me out every

body, it havent been that long that sprycel is on the market, so i dont beleive

doctors to much, because some say to take only tylenol then you go see another

I was given Tramadol 100mg for the bad times, and 50 mg for the so so times. If

her platlets are low it goes without saying no aspirin.

Did your doctor see something on your exams that made them think you

The prescribing information for Sprycel lists that 11% of people taking

the drug reported some weight gain. 1% reported a gain of more than

20% of their baseline weight before swithcing to Sprycel.

What dose of Sprycel did your Oncologist put you on?

Sorry to hear about the loss of PCRU...I switched from Gleevec to sprycel a

couple months ago..I had some headaches,fatigue,severe bone/joint/muscle pains

and spasms and severe constipation..Oh yeah..the big ol weight gain too..The

typical stuff I think..Doesn't seem to matter which med I take I have the same

damn side effects..lol..Just the way it is I suppose..I hope your transition

goes smoothly and regain PCRU..I check in two more weeks to see if I am

regaining my old levels before the switch..

Matt, please tell me there's more to the story than just losing your PCRU status

and please tell me that a clinical decision wasn't based on one single blood

test, more importantly, a test that wasn't even done in your regular lab.

You know that most of the experts don't believe PCRU should be a goal of

treatment right? A good stable response is what is hoped for by the top

experts.

I've gone in and out of PCRU more times than I can count. What matters is that

I remain stable within a one log range, not if a test is negative or positive.

In fact, to be honest, whenever I get a PCRU result I wonder if they screwed

something up. We all know that these drugs don't cure us and we know that we'll

always have cancer cells lingering around therefore, it's not comforting to me

to see a test that doesn't show any cells, it makes me think they didn't use

enough blood or run the sample long enough.

I've just seen too many examples of tests that bounced around to be concerned.

Unless there's a dramatic increase and by dramatic I mean OVER a log increase in

two or even three consecutive tests, I personally wouldn't make any therapeutic

changes.

Zavie,

I don't have that one(not yet anyways).Sure hope it is not something I do get

either..My DR does listen to my lungs at every visit.I have some shortness of

breath but I also have leaky heart valves and was told I would eventualy need to

be on oxygen..I'm hope not at 41 but maybe 81..lolI've only been on it for about

2 months now..Strugglin with losing the nearly 20 pounds in abouta month..Slowly

going down if I cut out all extras and count my calories closely..Hope it levels

off and then starts dropping..

My post wasn't clear..I gained nearly 20 pounds in the first month..Trying to

get that weight gain off..

Hi Sharon, last year on the xl228 drug I had severe constipation,  Nothing

seemed to help.

I discovered Kellogg's All Bran Bran Buds, They have 13 grams of fiber in 1/3

cup/  I don't like milk, but do like whipped yogurt, and that is my breakfast,

every day, I sprinkle the buds on the yogurt.  I also have it for snack in the

evening. And even though it is a bit high in calories, it " does the trick. "  

Ever listen to Dr. Oz on Oprah?  This is his terminology, not mine, but you

should " poop an " S " , every day.  I guarantee, if you eat these little all bran

buds, 1/3 twice a day, or 2/3 once a day, you will not have any problems.  Try

Sharon, sorry, I forgot to add, just to be sure, I also take a dose of Metamucil

every day.  I am going to stop the Metamucil, and see if just the Bran buds

work, no sense spending extra money if one doesn't have to!  Bobby

Bobby,

I am lactose intolerant so no dairy for me..I do have the All Bran Buds and

have used them but I don't eat them every single day..It hasn't helped me but I

will try them daily for a bit and see if that helps..I have had this issue for

10 years and tried just about everything everyone has ever suggested..Fact is I

have a very slow transit time..Takes days to move through..Maybe because I have

had WLS and my intestines have been cut and altered who knows..I will try it

daily and see if it helps..I can't take metamucil as it swells.I do use

benefiber as it doesn't swell and mirilax daily..Swelling is not good for those

who have had RNY surgery.Thanks

As far as the buds go, you can mix them with another cereal, and take them with

Silk, which is made of soy, right? I don't because I don't want to take the

milk, and can't even think of eating soyl..  But you must take them faithfully,

every day, to get 26 grams of fiber, and also drink lots of water.  By the way,

if you buy Miralax at Sam's club, you can buy two large ones for the price of

less than one at Walgreen's, just in case you didn't know, I think I paid under

10 for two large.  Always something, right?  Bobby
