Yes, QT Prolongation is the biggest

concern and that is why you need

weekly EKG's done for your first

month on the drug.

Over time, you body will adjust and

you will be better than ever. Just

wait, you will see.

to Glivec and I have been able to stop taking diuretics and other medication

I was taking for the migraines I was experiencing.

Dr Tim our leading CML specialist told me that if you were to eat a

fatty meal without fasting on Tasigna, that possible side effects could

affect the heart (QT prolongation)affect the heart (QT prolongation)<WB

experience issues with pancreas and rises in serum amylase, lipase etc. I

had a base line ECG and that was fine and my haem't is monitoring that

closely.

When you say " I can tell if I eat and then take Tasigna that it is more

potent, but not as much afterward " , what sort of side effects do you get?

Regarding your Tasigna question.

According to my oncologist, they want you to have an

empty stomach, so liquids or solids are supposed to

be kept to a minimum. It has to do with the absorption

and efficacy in your system, from what I understand. A

full stomach increases the potency and thus can trigger

heart or organ issues. For me, it seems that the no food

two hours before is the most important. I can tell if I eat

and then take Tasigna that it is more potent, but not as

much afterward. Just like Gleevec, you will find the things

that work and do not work for you. The standard things to

watch on Gleevec, you also need to watch on Tasigna

including Tylenol, St. s Wart and Grapefruit Juice, to

name just a few.

A lot of people find, especially after loose stools from

Gleevec, that they have constipation (a shock to your

system, especially if you have been on Gleevec for

several years) so I take my doses with a half of a glass

of Metamucil and I have no problems. Another thing to

watch out for is middle back pain. This may be a sign

of high Lipase, which is a concern of toxicity in your

pancreas, which I experienced early on as I made the

adjustment. You should call your Oncologist if this occurs.

One thing that I found on Tasigna was that I could drink,

which I really could not on Gleevec. So maybe you have

that to look forward to.

tonight, working my way up to 400mg. After the very unlovely side effects of

Glivec, I am hesitant about going up to " full " strength without a lead in

head cold I have had in years, which was developing before I started taking

Just a question for the more experienced - when fasting at night, can you

still have water? I hate to think what it will be like come summer if I

can't have any water for 3 hours!

I was going to start last night, then forgot to take the capsule at 7pm, so

as we had family around to celebrate my daughter's birthday, I didn't take

it last night. I can see I am going to need to be very disciplined, which I

am not as I would often forget to take my Glivec at teatime and take later

in the night.

You are welcome, but hopefully others

who have more experience than I am will

way in as well.

My first dose is when I get up in the morning

so my stomach is very empty then, it is the

second dose that is always tricky. I am

usually working, so I do not have a chance

to snack in the afternoon, so my second

dose is usually around five, which puts me

at least an hour before dinner.

While it is not a problem waiting an hour

to eat, sometimes I find I do need to eat

at the one hour point, or I start to feel bad,

but it may not be that way for you. Usually

five or six saltines, or Wheat Things (my

choice) are good.

Hi ,

I have been on Tasigna now for a little over 5 months. 157 days to be exact.

I keep track on my calendar.

I, like you had unlovely side effects from glivec and then with sprycel. So

now I am on Tasigna. When I started I started on 400mg., because like you I

was so afraid of side effects. My PCR was 0.011 at the time and that was

because I was off all meds for almost a month prior to starting Tasigna. I

had absolutely no side effects with Tasigna. Maybe a little weight gain but

I contribute that to overeating which maybe the Tasigna made me have an over

active appitite. Anyway, this is what I decided to do. I get up almost every

morning to go to the bathroom anyway-about 5am. So I have my pills in the

bathroom and I take the two 200mg pills at that time and go back to bed so

that when I get up at 7am, I can have my coffee. I love my morning coffee.

So then I went for another PCR about a month ago and it was 0.032. so they

increased my Tasigna another 200mg. and this one I take about 5pm. I try to

remember to stop eating at 3pm so that my stomach is empty at 5pm when I

take the pill. Then I do not eat until 7pm. The rule, that I understand,

is take Tasigna on an empty stomach 1 hour before you eat or two hours

after. But to be safe I do it 2 hours before I eat.

So I have been on 600mg for a month and today I go in for another PCR to see

how I am doing. I am hoping that the results are good. If not they will have

me take another 200mg.

About water: I feel yes you can have water-you take the pill with water so

I am sure it is ok to drink water. They say FOOD. Although I would not drink

anything else.

I have no side effects on 600mg.

Nice to hear fron another person on Tasigna.

I was going to start last night, then forgot to take the capsule at 7pm, so

as we had family around to celebrate my daughter's birthday, I didn't take

it last night. I can see I am going to need to be very disciplined, which I

am not as I would often forget to take my Glivec at teatime and take later

in the night.

Hi Matt and Bobby,

I do not participate much verbally to this site but I do read most of the

posts.

It is always good to hear from other Tasigna people.

I have tolerated my 600mg very well. 400 in the morning and 2 in the

evening.

Today I have an appointment for a PCR to see how I am doing on 600mg. I hope

well.

Are both of you PCRU???? I hope so.

My last test a month ago showed me at 0.032-up a little

But so many of you out there said that was not terrible and not to worry.

I do not know what I would do without all of you.

But I must say --- on 600mg---I feel great. No side effects except for

weight gain. But I think that is my fault. Not Tasigna. I am only 5 ft and

every little pound shows on me.

My first dose is when I get up in the morning

so my stomach is very empty then, it is the

second dose that is always tricky. I am

usually working, so I do not have a chance

to snack in the afternoon, so my second

dose is usually around five, which puts me

at least an hour before dinner.

While it is not a problem waiting an hour

to eat, sometimes I find I do need to eat

at the one hour point, or I start to feel bad,

but it may not be that way for you. Usually

five or six saltines, or Wheat Things (my

choice) are good.

Yes, QT Prolongation is the biggest

concern and that is why you need

weekly EKG's done for your first

I was taking for the migraines I was experiencing.

Dr Tim our leading CML specialist told me that if you were to eat a

fatty meal without fasting on Tasigna, that possible side effects could

affect the heart (QT prolongation)affect the heart (QT prolongation)<WBR<WB

experience issues with pancreas and rises in serum amylase, lipase etc. I

had a base line ECG and that was fine and my haem't is monitoring that

closely.

When you say " I can tell if I eat and then take Tasigna that it is more

potent, but not as much afterward " , what sort of side effects do you get?

Regarding your Tasigna question.

According to my oncologist, they want you to have an

empty stomach, so liquids or solids are supposed to

be kept to a minimum. It has to do with the absorption

and efficacy in your system, from what I understand. A

full stomach increases the potency and thus can trigger

heart or organ issues. For me, it seems that the no food

two hours before is the most important. I can tell if I eat

and then take Tasigna that it is more potent, but not as

much afterward. Just like Gleevec, you will find the things

that work and do not work for you. The standard things to

watch on Gleevec, you also need to watch on Tasigna

including Tylenol, St. s Wart and Grapefruit Juice, to

name just a few.

A lot of people find, especially after loose stools from

Gleevec, that they have constipation (a shock to your

system, especially if you have been on Gleevec for

several years) so I take my doses with a half of a glass

of Metamucil and I have no problems. Another thing to

watch out for is middle back pain. This may be a sign

of high Lipase, which is a concern of toxicity in your

pancreas, which I experienced early on as I made the

adjustment. You should call your Oncologist if this occurs.

One thing that I found on Tasigna was that I could drink,

which I really could not on Gleevec. So maybe you have

that to look forward to.

Hi Sharon,

We can nearly have our own little club now LOL. With Tasigna I am taking it

at 6.30 -7am when I get up, w/e often 8am, then in pm, start fasting at 5pm,

take meds at 7pm and eat at 8pm. I guess if I need to could have a little

earlier. Will be good for the diet as I tend to pick a little when I get

home from work around 6pm, so will be a definite " no no " . I'm more concerned

I can't have a cup of tea for 3 hours!

It's been great giving the feedback from this group to my haem/t as he was

very receptive to commencing on the lower dose initially.

When I think of my first 10 days on Glivec - I wouldn't know I am even

taking anything with Tasigna....just wonderful!

I have been on Tasigna now for a little over 5 months. 157 days to be exact.

I keep track on my calendar.

I, like you had unlovely side effects from glivec and then with sprycel. So

now I am on Tasigna. When I started I started on 400mg., because like you I

was so afraid of side effects. My PCR was 0.011 at the time and that was

because I was off all meds for almost a month prior to starting Tasigna. I

had absolutely no side effects with Tasigna. Maybe a little weight gain but

I contribute that to overeating which maybe the Tasigna made me have an over

active appitite. Anyway, this is what I decided to do. I get up almost every

morning to go to the bathroom anyway-about 5am. So I have my pills in the

bathroom and I take the two 200mg pills at that time and go back to bed so

that when I get up at 7am, I can have my coffee. I love my morning coffee.

So then I went for another PCR about a month ago and it was 0.032. so they

increased my Tasigna another 200mg. and this one I take about 5pm. I try to

remember to stop eating at 3pm so that my stomach is empty at 5pm when I

take the pill. Then I do not eat until 7pm. The rule, that I understand,

is take Tasigna on an empty stomach 1 hour before you eat or two hours

after. But to be safe I do it 2 hours before I eat.

So I have been on 600mg for a month and today I go in for another PCR to see

how I am doing. I am hoping that the results are good. If not they will have

me take another 200mg.

About water: I feel yes you can have water-you take the pill with water so

I am sure it is ok to drink water. They say FOOD. Although I would not drink

anything else.

I have no side effects on 600mg.

Nice to hear fron another person on Tasigna.

Hi everyone,

I commenced Tasigna last week - 400mg once a day and will commence 200mg

tonight, working my way up to 400mg. After the very unlovely side effects of

Glivec, I am hesitant about going up to " full " strength without a lead in

and my haem/t has been very supportive of this.

So far I have tolerated 400mg with no problems, except that I have the worse

head cold I have had in years, which was developing before I started taking

Tasigna.

Just a question for the more experienced - when fasting at night, can you

still have water? I hate to think what it will be like come summer if I

can't have any water for 3 hours!

I was going to start last night, then forgot to take the capsule at 7pm, so

as we had family around to celebrate my daughter's birthday, I didn't take

it last night. I can see I am going to need to be very disciplined, which I

am not as I would often forget to take my Glivec at teatime and take later

in the night.
