Both came back 1100 - Not a significant level to cause the side effects I had. I had a good response to gleevec reaching PCRU in 9 months and holding it since. I think to test inside the cells would be more accurate. Guest guest Posted July 17, 2008 Report Share Posted July 17, 2008 G’day to the 2 Barb’s, I had a PK level done back in June as due to severe side effects of Glivec 400mg, I had a break from Glivec for 6 weeks, then have been on 200mg since. My PK level was only 480ng, which is obviously too low, but on 200mg I feel great! It would have been interesting to see what it was when I was on 400mg. I am having monthly PCRs at present, going from 13.01% (Jan) to 0.08% (April), then 0.02% (May) after the break and last one was back to 0.09% (June). Still waiting for my current results, it’s a pain having to wait 3 weeks or more for them. We only have one lab in Australia where the test can be done and fortunately it is only a 5 ½ hr drive, and I have a friend who lives there so a good “girls” w/e away! Tim who is the leading specialist in CML in OZ, told me that it is possible 200mg is fine for me, but as I was only dx last July – gee my first anniversary is only a week away, he doesn’t want to “risk” me developing resistance or mutations, so will be swapping me over to Tasigna. Tim made the comment at this stage that testing cannot be done at cellular level – this would be great for me as would prove whether or not 480ng is a therapeutic dose. But they are working on it – we have the PK test, so who knows what they can do within the next few years? Guest guest Posted July 18, 2008 Report Share Posted July 18, 2008 I had the test done in 2007 as I was not getting better than 2ish log reduction for over two years, and I had a plasma level of 600ng, which is below the optimal 1000 but now my PCR is to low to get a number, although it still is positive (-4 or 5 log reduction), and I stayed at 400mg of gleevec a day. The test was done by the lab in Toulouse France, even if I live in California (the blood got shipped at Novartis expense). Yes there is a large variance of results around the 1000ng optimal value. My dr at Saint Louis hospital in Paris, who did the referral for the test, told me above 500ng is acceptable as long as the PCR results are good. Marcos.
