However, it isn't just the amount, it's also a matter of when to take it. I take all 100 mg in the morning which is what is recommended. I did have one bout with pleural effusion early on but that was taken care of by going off Sprycel for 3 weeks and tasking 40 mg of lasix per day. I now take a maintenance dose of 20 mg lasix and don't have a problem. I did have about 30 pounds weight gain on Sprycel which was also a problem on Gleevec until I lost my response to that drug and lost all the weight, too. I've been on Sprycel in a clinical trial at the University of land since early 2004. When I was first diagnosed, I wanted the highest dose I could stand, but Tracey helped me see that a lower dose with fewer side effects was better for your quality of life. What I meant to say is to take the full dosage regardless of what time of day you take it. (When a was put on 70mg, she was in a trial of escalating doses and then just stayed there because it was effective for her). Sprycel does not actually have a therapeutic dose like Gleevec does. Because Sprycel is so much stronger, some people are on 40-50mg and it works for them.....but the starting dose now is 100mg. If I were you, I would go up to 100mg, see if you get your old pcru status back....and then if your side effects are bad, see if you can drop the dose and still maintain. That is what I see the specialists doing from my end. I am a Dr. Druker patient and I am in a Sprycel trial, and started at 100mg for 3 years....then had some pleural effusion and we have gone back and forth between 80 and 100mg because of PE (and I may end up switching to Tasigna) because the 80mg dose is not really therapeutic for me. Last April, I was switched from 600mg of Gleevec to 100mg of Sprycel and within 6 months, I was in remission! Whoo hoo! It was also never recommended to me to take the medication in the morning, just as long as you take it the same time every day. I take mine in the evening also usually when I have dinner (that’s helps me remember ) From the medication, I have minimal side effects to what I had with Gleevec but I have seen a tremendous weight increase on Sprycel when I was losing while on Gleevec. Out of curiosity, has anyone experienced a difference in their skin pigment since taking Sprycel? I’m half Italian so I could be in the sun for hours upon hours and never burn but ever since I started, even an hour in the sun burns me like a tomato! Not quiet as dark any more But I have learned if I get a little sun at a time I tan and not burn. Yes I have burned in the beginning days of chemo until I learned to treat my skin as if I was fare complected. By getting small amts several times a day i was able to build up and able to be in the sun all day without burning. I am under the impression that the chemo affects the melatonin. Guest guest Posted January 20, 2010 Report Share Posted January 20, 2010 Matt, After diagnosis in March 2007 and after discovering I couldn't tolerate Gleevic I was put on Sprycel at 100mg daily. It didn't take long before I was experiencing breathing problems and extreme fatigue. I was lowered to 50 mg daily and slowly tried to increase and got to 70 mg when I once again had to take a medication vacation because of side effects. I am now taking 50 mg every other day and maintaining a great response. I have heard of some that are on as low as 20 mg daily. I should add that Sprycel got me to PCRU within 3 months and I have managed to stay at PCRU or at a trace but unquantative since October '07. When I went to MD in July '09 and met with Dr. Cortez he said the key is to find the lowest dose that will still keep you at a major response and also provides a good quality of life. Everyone is different but there is nothing that says you must stay at 100 mg.
