Abstract

The mechanisms of cell toxicity of mycotoxins of the enniatin family

produced by Fusarium sp. enniatin B, a mixture of enniatin homologues (3% A,

20%

A(1), 19% B, 54% B1) and beauvericin, were investigated. In isolated rat

liver mitochondria, exposure to submicromolar concentrations of the enniatin

mycotoxins depleted the mitochondrial transmembrane potential, uncoupled

oxidative phosphorylation, induced mitochondrial swelling and decreased

calcium retention capacity of the mitochondria. The mitochondrial effects were

strongly connected with the potassium (K(+)) ionophoric activity of the

enniatins. The observed enniatins induced K(+) uptake by mitochondria. This

shows that the enniatins acted as ionophores highly selective for potassium

ions. The effects were observed in potassium containing media whereas less

or no effect remained to be observed when K(+) was partially or totally

replaced by isomolar concentrations of Na(+). The rank order of enniatin

These tests were performed on mitochondria isolated from cells and tests were

then performed in test tubes. Mitochondria are the cellular organelles that

utilized oxygen we breathe and then converts the use of the oxygen into energy

producing chemicals called adenosine triphosphate (ATP). ATP is the energy

source to metabolize organic compounds we consume as food and cert them into

proteins, DNA, RNA and by-products excreted in the urine, feces and bile.

Abscence of the function of mitochondria leads to chronic fatigue and eventually

death.

The only problem with this study is that it was done in a test tube on isolated

mitochondria. We do not know for sure what occurs in the living cell. A study

such as this sets up future studies on whole living cells.

Dr Thrasher,

Are the species and resultant toxins tested in this study generated in damp

buildings or food? Fusarium is more often associated with moldy food.

However, it raises the question do other toxins from WDB have a similar effect

on mitochondria?

Carl: I am searching that question presently. I have not found a good

reference on Fusarium species in buildings. Do you have one? At present, the

mycotoxins in the study are most likely foods, e.g. grains of various types.

Carl: I have completed my search for Fusarium species indoors. The papers only

list genus. Therefore, what species are indoors are unknown. There are some

papers on nosocomial infections and species involved.
