Amoxicillin-induced flair/effect on HHV6 replication

Abstract

Amoxicillin is known to induce exanthema in patients with EBV-induced infectious

mononucleosis. It is widely recognized that the reactivation of herpesviruses,

including HHV-6 (Human Herpesvirus 6) and EBV (Epstein Barr virus) is associated

with DRESS (Drug Reaction with Eosinophilia and Systemic Symptoms). We report 7

cases of amoxicillin-induced flare in patients with DRESS induced by other drugs

and investigate whether amoxicillin may have a direct effect on HHV-6

replication in vitro. 7 cases of DRESS with amoxicillin-induced flare were

retrospectively analysed. The influence of amoxicillin on HHV-6 HST strain

replication was studied in vitro in a human T lymphoblastoid MT4 cell line. The

viral replication was quantified by immunofluorescence assay and by real-time

polymerase chain reaction (PCR). Comparisons were performed using the Student's

t test. Amoxicillin-induced flare was observed in 7 patients with DRESS induced

by other drugs. In two cases HHV-6 reactivation was studied and was demonstrated

by PCR. Amoxicillin neither modified cell viability nor cell proliferation for

the range of tested concentrations.

Amoxicillin increased the replication of HHV-6 at 25 microg*mL-1 and 50

microg*mL-1. Amoxicillin may induce a flare of DRESS, possibly by acting

directly on herpesvirus replication.

My son regressed severly into autism, we lost him while on Augmentin. I always

wondered why, now I guess I know. I plan to bring this up when we visit a

doc this spring.

My son took Amoxicillin often as an infant and toddler. He was having some mild

delays until age 4 when he took amoxicillin for an ear infection and he totally

spiraled down into autism for 3 days. Lost eye contact, wouldn't play with

toys, just sat and spinned around in circles.

I took him off the Amoxicillin and tried to figure out if it was a reaction to

the med, but our doctor just didn't believe me. The doctor didn't t think

amoxicillin caused any of his problems. He got a little better after getting

off it, but never got back to where he was before the reaction.

He is 10 years old now and I am more confused than ever. We are trying ,

but not seeing much improvement after 7 months. He is diagnosed with PDD-NOS,

and Childhood Distintegrative Disorder. He has many symptoms of autism, but

isn't like any other " autistic " kid I know. He has so many delays. Very low

large motor skills, and hardly any fine motor skills. Hardly any communication

skills...No clue what the word " no " means.

Is there any possibility that he was exposed to a particular bacteria that

triggered his spiral?  Has he been properly screened/tested for Lyme and co

infections.  My daughter has Lyme and Bartonella which affects her CNS and

function.  She is taking zythromax and bactrim now and making

improvements...she

is worse with Amoxicillin.  Just a thought.
