I started him on oral B12 with 5MTHF pill (mixed 1/2 cap with almond milk) in the morning over the weekend.So far no hyperactivity immediately afterwards. But he's been really "out of it" attention wise overall...less language use, and also he's started lining up his legos! Something he hasn't done since he was 2.5 yrs old. I'm worried. Then just after dinner he was running around like a maniac and generally behaving crazy. This morning again, out of it when he woke up, less language and scratching himself all over his torso until I put his clothes on. He was quite out of it overall again and did the lining up with legos again :( My biggest fear is a permanent regression. What do you make of this? Could it be yeast?Is there a relationship between MB12 / B12 and yeast?My child has a methylation issue...one copy of the C677 mutation. This is a negative reaction and I really think that negative reactions are better than no reaction at all bc now this is a clue to the pathway you should work on. You mentioned one wheel in the methylation pathway, but how are the other wheels? I'm thinking more specifically about the BH4-BH2 wheel or the methionine wheel. Are you doing an adequate amount of b6/p5p? That would affect the neurotransmitter production and transsulfation components. Why I think it's the BH4-BH2 wheel is bc that affects serotonin and dopamine. Also, if you give b6, you might want to add some magnesium w that. I would suggest mag taurate to help out the transsulfation too, I think. Currently my child is on taurine and also Vit D, C, spectrum complete, enzymes, nystatin, L carnitine. We haven't given B6 or P5P yet.....there's just too much I need to put in his mouth. We have just added B12 with 5-MTHF...so that's where we are at right now. Do you think I should add B6/p5p now?Thanks for your help. When you work on methylation, it's often helpful to feed other parts of the methylation pathway. I think where your son might be affected is with the BH4-BH2, where it produces serotonin and dopamine (which dopamine goes on to produce epinephrine and norepinephrine). It really sounds like he's having some trouble feeling at ease, hence the need for the hyperactivity behaviors. If you run an OAT, you might see that his serotonin is low and it'll probably tell you to supplement 5-HTP. You want to watch the clostridia markers and you don't want to do tyrosine or phenylalanine if you have high clostridia markers because it'll just feed the clostridia and they'll just oxidize the dopamine. But 5-HTP might be helpful. Just sayin...you might want to watch the other wheels in the methylation pathway if he's having a negative reaction to folate and you might want to run some tests and maybe check out those other wheels. So I usually increase my yeast fighter (candex, grapfruitseed extract)when my son is on the B12 shots. Guest guest Posted May 21, 2012 Report Share Posted May 21, 2012 We never did the shots bc I was worried about over absorption and cobalt toxicity at the time. We did sublingual and I didn't see anything like that with my daughter but my son developed some of that behavior after a bit of the sublingual so I stopped it. We ran some bloodwork and it turns out he's got very high b12 even several months without supplementation and our DAN said that prob not enough folate for him to get the b12 into his CNS. Since then we've been working on my daughter's folate and mito support so I might try the shots after a few more rounds of chelation. In retrospect, it could have been related to his CBS mutation where adding MB12 may have resulted in more ammonia buildup.
