If you use a proper starter each time, you can be absolutely sure that there is a minimum of contamination each time you make it. I concur with Mara plus it's written explicitly in BTVC. That said I did exactly what you're asking last week (due to running out of starter) and I found the 2nd generation yogurt to be significantly different than 1st gen.
